Hi everyone.  I am newly diagnosed stage 4 and was wondering if anyone has participated in a trial that combines taxol with reparixin?  It's fairly new, randomized and in phase 2.  I was wondering how you are responding and any side effects.  Thank you!

I'm sorry you are dealing with a recurrence.  Where did it metastasize?  I don't have any information on this study, but there was this release of the results of hte Phase 1B study.

Reparixin is a CXCR1/2 inhibitor with a low molecular weight that in experimental studies has been shown to be able to reduce cancer stem cells if administered alone or in association with chemotherapy with medicines of the taxane family. With reparixin, the aim is to inhibit the activity of the "driving force" of the inflammation in order to keep under control the development of breast cancer stem cells and, potentially, other neoplasia as well, in addition to altering the microenvironment that allows their development.

Other immunotherapy approaches

“Our immunotherapy studies have moved from just vaccines to include checkpoint inhibitors and other immunotherapy strategies,” Dr. Mittendorf said. “And as an institution, we have the opportunity to do more.”

For example, Dr. Ibrahim is the principal investigator at MD Anderson for an ongoing multi-institutional trial of the oral immunostimulant indoximod with taxane-based chemotherapy for patients with HER2-negative metastatic breast cancer. Indoximod inhibits the indoleamine 2,3-dioxygenase pathway, which suppresses T cell activity.

Also under investigation are agents that block the action of interleukin-8 (CXCL8), which has an immunomodulatory effect and is associated with breast cancer growth and metastasis. A trial of one such agent, reparixin, which binds to the CXCL8 receptor CXCR1, is underway at MD Anderson and other institutions.

Preclinical studies of promising therapies also are underway. For example, Dr. Mittendorf said, Laurence Cooper, M.D., Ph.D., a professor in the Division of Pediatrics, is investigating the use of T cells modified with chimeric antigen receptors (see “Sleeping Beauty” Technique Modifies T Cells to Treat B Cell MalignanciesOncoLog, May 2014) against triple-negative breast cancer.

Just wanted to post an update if anyone reads this.  After the first cycle of 2 months, my results did show that the tumor has shrunk from 13 x 10 mm to 11 x 7 mm!

IDC, 2.2 cm, Stage IIb,lumpectomy 1/30/09 ACx4,Tx4 36 rads
6/1/16 Local recurrence same breast, same spot 1.8cm Carb.4x every 3 wks, Taxol 12x once wk. Dbl Mast. PCR!! Reconstruction fail, NED!

I'm doing fine...maybe decent is a better word. I think most of my side effects sten from the taxol--just the usual stuff--hair loss, neuropathy (not too bad), weight gain. Sometimes I'm a little gassy right after I take the study drug, but that's it.
